I couldn't agree more with Bush Baptist that all road in the CB-area can produce stunning sightings!
Talk to people and look at the sightings board and then decide where to go.

I had very good sightings on the S25 also later on during the day (particularly in winter). Where the S25 crosses the riverbeds of the Bume and later on the Biyiamiti may produce good sighting throughout the day. In winter you may consinder to drive to Gayisenga (S108, saw wilddogs there in the dry riverbed) and Mpondo dam (102, predator hotspot in winter when the surounding areas can be very dry).

Lindanda, Salitji, S100, Kanniedood dam, Mahonie loop.

The S25 seems to be the most popular.

In terms of famous roads I think the following roads fall in that category:

Lower-Sabie to Skukuza,
Crocodile Bridge to Lower-Sabie
Orpen to Satara
The historical Voortrekker Road and
Letaba to Satara

I think Grantmissy is on it.
The five most famous will be the five busiest roads.
Skukuza-Lower Sabie H4-1
Lower Sabi-Croc Bridge H4-2
Malalane-Skukuza H3
Phabeni-Skukuza S1
S100

but those are not necessarily the best in Kruger
